银屑病皮肤白细胞介素8的免疫组织化学研究

An immunohistochemical study of interleukin-8 on the psoriatic skin

应用抗白细胞介素8(IL—8)的单克隆和多克隆抗体对IL—8在银屑病皮肤的定位与分布进行了免疫组织化学研究.进行期寻常型银屑病患者和健康对照各7例,分别切取病损处及其周围未受累皮肤和正常人皮肤进行冰冻切片.用IL—8抗血清和两种IL—8单抗(5A4、2G2)以ABC免疫组化技术染色观察结果.发现IL—8抗血清和单抗5A4能使正常人表皮着色,且基底细胞层明显深染.5A4、2G2在银屑病皮损表皮的基底以上层呈现阳性,而未受累皮肤的染色结果无明显规律性,银屑病皮损表皮与正常人皮肤IL—8的分布差异表明IL—8在该病的病理生理过程中可能发挥重要作用.

Intetjeukin-8(IL-8)is a potent 8KD proinflammatory peptide that has been purified from posriatic scales.Its chemoinctic activity on human neutrophils,and its presence in psoriacic skin may have some redlationa to this discase.Inthe present study,IL-8immunoreactivity was detected in normal and psoriatic epodermis using polyclonal antiserum and two different MoAb(5A4,2G2)against IL-8.It was detected in both notmal and paoriatic skin,and was most intedse in the baas cell layet of normal skin with antisetum and 5A4,and there was ataing of psoriacic damagcd skin with 5A4 and 2G2 above the basal cell layet.Irregular positive reaction was found in the psoriatic non-damaged lkin with antisetum,5A4 and 2G2.The sweat glands and the hair follicels were IL-8 immunoreactive in all biopsies studied.The findings revealed the existance of IL-8 in both normal end pscriatic skin ,although its localization was different.Furthet inveatigations are needed to learn ist role in the pathogenesis of psoriasis.
