摘要
建立单光子辐射方式的液闪技术化学发光(LSC-CL),灵敏、简便、自动测试。以多型核白细胞(PMN)为模型,探讨了基础发光、依赖发光、最大吞噬发光,显示了较好的峰形(组间P<0.01),对方法学进行因素水平选择,正交[L_9(3 ̄4)]设计,最佳因素和因素水平序为:(1)LS阀值I(0.5)、(2)酵母多糖调理Ⅱ(1:0.5)、(3)pH值Ⅱ(7.6)、(4)Luminol水平Ⅱ(10 ̄(-4)M)。临床用于老年慢性支气管炎和儿童支气管肺炎极期PMN一CL的测定,结果表明:极期PMN吞噬功能低下(P<0.0l),老年组更低。提示PMN一CL的检测,对临床研究抗感染机制和自由基损伤机制具有重要意义。
The liquid scintillator counting-chemiluminescence(LSC-CL)of monophoton radiance is a sensitive,handy ,high-autoanalytic technique.Though measuring basic CL,dependent CL and maximun phagocyticCL of polymorphonuclear (pMN),we studied best factor levels of the metod with orthogonal dessign [L_9(3 ̄4)]. The results showed the peak forms changed markedly P<0.01),and the order of opti-mum factor levevls :(1)Ls threshold I(0.5),(2) zymosan A opsonize Ⅱ(1:0.5),(3)pHII(7.6),(4)theIuminol nevel III(10 ̄(-4)M).It was measured that PMN-CL in blood during acute attack of the old patientswith chronic bronchitis and the childem with paeumonia bronchial .It was suggested that PMN phagocytosisdecreased.So the dynamic analysis of maximum phagocytic CL would help us with the deepgoing clinical re-searches of the mechanisms of anti-inflammation and injured by the oxygen free radicals.(
