摘要
研究了不同培养基及外植体条件下麝香石竹(Dianthusfragrans)玻璃苗的诱导形成及玻璃苗部分生理特性。结果表明,提高培养基琼脂浓度或采用较大外植体,玻璃苗出现频率降低,以幼小茎尖为外值体或降低琼脂浓度,则玻璃苗出现频率增加。玻璃苗叶绿素、蛋白质含量及呼吸和光合速率低于正常苗,含水量、相对含水量及组织自由水含量显著高于正常苗,而束缚水含量低于正常苗。本文研究认为:麝香石竹玻璃苗的形成是培养基和外植体因素相互作用的结果。培养基极端的水分环境可能是导致玻璃苗形成的外在原因。
A study was conducted on vitreous shoot formation under different conditions of media and explant. and physiological characteristics of vitreous shoot in carnation (Dianthus fragrans). Large explants or culture media with high agar concentration reduced the percentage of vitreous shoot,and small tip buds or media with low concentration of agar raised it. As compared with normal shoot, the chlorophyll and protein contents,the respiration and photosynthesis rates,the percentage of bound water of vitreous shoot lowered obviously: but the total water content, the relative water content and percentage of free water rose. The value of relative water content of vitreous shoot rose above 100%. It is clear that the water content of vitreous shoot tissue is excessive.The results of the study showed that the vitreous shoot formation is related not only to the media factors, but also to the explant itself. The extreme water condition of media is probably the external cause inducing vitreous shoot formation.
