摘要
采用增生细胞核抗原(PCNA),Ki-67和溴脱氧尿啶嘧核苷(BudR)标记的S期细胞的三种生物学指标,测定人食管和贲门粘膜活检上皮正常和不同程度的癌前病变细胞增生状况。食管粘膜上皮从正常到基底细胞增生,上述指标变化幅度不大,但从基底细胞增生到间变时,PCNA和Ki-67阳性细胞数明显升高。贲门随上皮病变的加重,PCNA和Ki-67阳性细胞逐渐升高。在同一类病变,PCNA和Ki-67阳性细胞数几乎是BudR的2倍。结果提示三种指标均与食管和贲门上皮病变进展程度有关,能比较客观地评价其细胞增生动力学状况。PCNA可能是一种简便易行,特别适宜研究大范围人群细胞增生状况的生物学指标。
The main objective of this study is to assess the cell kinetics of human esophageal and gastric cardial biopsy of the epithelium with normal and different severities of precancerous lesions by measuring the proliferating cell nuclear antigen (PCNA), Ki-67, and bromodeoxyuridine (BudR) incorporation. The samples were from an area of high incidence of esophageal cancer in Linxian and Huixian, Henan Province. In esophageal biopsies, these biomarkers changed slightly between normal and basal cell hyperplasia(BCH),but increased greatly between BCH and dysplasia. In cardial biopsies, the number of PCNA and Ki-67 positive cells increased progressively from normal chronic superficial gastritis (CSG) ,chronic atrophic gastritis (CAG) and dysplasia. The positive cell number of PCNA and Ki-67 was almost two times higher than that of BudR in the same category of lesions. PCNA may be an promising and simple biomarker to assess cell proliferation,especially in the large scale study of population.
