高温热疗引起小鼠Ehrlich腹水癌细胞凋谢的研究

摘要 用银染核仁形成区(AgNORs)及酸性DNA酶染色法,对不同高温处理的小鼠Ehrlich腹水癌细胞进行光、电镜观察。结果显示高温(41℃~43℃)处理可使癌细胞以典型的凋谢(Apoptosis)方式死亡。凋谢出现率比37℃孵育组升高,且43℃孵育组的凋谢出现率高于41℃组(P<0.01)。高温处理组,癌细胞的核仁形成区银颗粒数低于37℃孵育组(P<0.01)。凋谢细胞和凋谢小体的银颗粒数也比其它癌细胞为低。高温处理引起的凋谢,细胞核及凋谢小体核碎片酸性DNA酶染色呈阳性反应,与其它细胞核相比明显增强。结果提示,肿瘤高温热疗的机理之一是启动了癌细胞的凋谢过程,直接损伤肿瘤细胞;整体热疗也可能通过刺激免疫反应等间接作用而达治疗作用。 The change of Ehrlich ascites cancer cells incubated in 41℃ and 43 ℃ hyperthermia was observed by means of light,electron microscope and acidic DNAse. The results showed that hyperthermia (41℃ and 43 ℃) induced the incidence of apoptosis of the cancer cells of the ascites. Percentage of apoptosis in 41 ℃ or 43 ℃ incubation was higher than that in 37 ℃ incubation;the AgNORs number in the cancer cell nuclei after hyperthermia (41 ℃ and 43 ℃) treated was lower than that in treated with 37 ℃ incubation and AgNORs number of apoptotic also lower than that of the intact nuclei. There was intense activity for acidic DNAse in the nuclei of apoptotic bodies. The result suggested that one of the mechanisms of hyperthermia therapy may start the process of apoptosis of cancer cells. The paper also reports that mild hyperthermia may have a treatment effect by irritating immunity reaction of the body.
