摘要
文章选用了二种酯化条件,将L-3,5,3’,5’-四碘甲腺氨酸酯化成L-3,5,3’,5’-四碘甲腺氨酸甲酯。用此甲酯半抗原和牛血清白蛋白偶联,合成了甲状腺素免疫原。经免疫兔子获得了抗体。抗体的滴度为1:3200—14000,亲和常数为3.5×10~9l^2·mol^(-1),在L-3,5,3’-三碘甲腺氨酸过量L-3,5,3’,5’-四碘甲腺氨酸碘标记物100—1000倍时交叉反应值>0.0001。
Thytoxine (T_4) is esterified to T_4-methylester under two different conditions.Then T_4-methylester is conjugated with bovine serum albumin. The conjugate of T_4-methylester with bovine serum albumin is injected into rabbit's bodies to obtain a highly specific antibody for T_4. The titer of T_4-antibody is 1:3200-14000, the high affinity constant is 3. 5×10~9 1~·mol^(-1), and cross-reactivity for T_3 is less than 0.0001 (as T_3 is 100-1000 times of labeled T_4). The T_4-antibody is used for the production of Thyroxine RIA Kits for quite a few years with good records.
