摘要
文章介绍了以17β-雌二醇(17β-E_2)为底物,合成制备雌二醇-6-肟-组胺,用^(125)I碘化后,经一步硅胶GF_(254)薄板层析(TLC)分离,即可得到^(125)I-雌二醇-6-肟-碘代组胺的方法。用自身置换法测定比活度约74MBq/μg。并对其免疫活性进行了实验。与过量抗体(10倍)结合率平均82.5%(n=3)。认为是建立17β-雌二醇放射免疫分析方法比较理想的试剂。
Using oestradiol-17β as a starting material, oestradiol-6-oxime-histamine and^(125)I-oestradiol-6-oxime-iodohis tamine are synthetized. ^(125)I-oestradiol-6-oxime-iodohistamine purified by thin layer chromatography (T.L.C)plate, at a specificactivity of about 74 MBq/μg determined by selfdisplacement method, shows a highstability and a fine immunoreactivity tested by excess antibody binding. Suggestingits suitability as tracer for radioimmunoassay (RIA).
