摘要
用改良Percoll梯度柱离心分离牛精子,并将分离后富含X精子的底层精液进行配种实验。结果发现,底层精子活率正常,含F小体的Y精子仅10.2%。将其鲜精直接配种母牛13头;冻精配种81头。鲜精组产仔率84.6%,冻精组为72.83%,与对照组比较无差异。母犊出生率,鲜精组为81.80%,冻精组为71.18%,分别比对照组提高36.38%与25.78%(P<0.01)。结果表明,此分离方法可提供富含X精子的精液;应用于人工配种。
eparation of bull X-and Y-bearing sperm by the Percoll density gradient
centrifugationand the succession animal test with the X sperm-rich fraction were described in
this paper. After centrifugation,the sperm was distributed on the interface with discontinuous
Percolldensity gradient.Samples form each interface were collected and their purity of the
separa-tion was examined by meams of F-body.The content of the F-body sperm in the bottom
ofthe tube was only 10.2±2.6%,meanwhile,the motility of the separation sperm was
nor-mal.Artificial insemination was performed in 13 heads of milk cow,using the fresh bottomX
sperm-rich fraction and in 81 heads of milk cow,using the freezing bottom X sperm-richfraction.
The pregnant rate was 84.6%,and 72.83%,respectively,which was the same asthe control group.
The birthrate of female calf was as high as 81.%,in fresh sperm group,71.18%in freezing sperm
group.an increase of 36.38%,25.78%respectively,in comparisonwith the control group.In the
development milk cow enterprise,the method of Percoll densi-ty gradient centrifugation could
provide X sperm without loss of its motility.
