摘要
研究Bouin液、10%福尔马林和4%多聚甲醛与0.25%戊二醛混合固定液(简称混合固定剂)及不同固定时间对降钙素(CT)免疫组化显示成年大鼠肺神经内分泌细胞(PNEC)的影响。计量分析发现Bouin液保存CT抗原性的效果最好,但受固定时间影响较大,以固定4h效果最好。混合固定剂保存CT抗原性的效果最差。
mmunohstochemistry is a powerful tool for analysing sparsely distributed pulmonaryneuroendocrine cells(PNEC).The technique has afforded sensitive and specific detection ofPNEC’s peptides and amines. However,fixatives and fixation time condition vary in theirsuitability for immunoreactivity. The authors examined the effects of 3 kinds of fixatives and3 different fixation times on adult rat calcitonin(CT)immunoreactive PNEC,and measuredthe mean number of CT positive cells per cm2 section area,The results were as follows:(1)Bouin was the best fixative. This fixative could demonstrate solitary neuroendocrine cells(NEC)and neuroepithelial bodies(NEB)clearly,but it was influenced by fixation times.Specimens treated with Bouin for 4 hours were found to be best suited for immunohis-tochemistry.(2)Tissues fixed with 10%formalin and mixed fixative including 4%paraformaldehyde and 0.25%glutaraldehyde was less desirable for immunohistochen1istry.These 2 kinds of fixativeswere only demonstrated NEC and small aggregates of NEB,andnot influenced by fixation times.
