摘要
用Percoll密度梯度离心法从志愿者血液中分离出感染间日疟原虫的红细胞。以此作为免疫原,免疫Balb/d小鼠,获得5株杂交瘤细胞系。融合率平均为45%,抗体阳性率为0.66%。用IFAfoIPA染色进行免疫组化分析,4B2和8E3分泌的单克隆抗体呈较强的阳性反应,与约氏疟原虫红内期无交叉反应。
With Percoll
density gradient centrifugation, the erythrocytes infected P. vivax fromvolunteers were
separated as immunogen and immunized Bale/c mice,in which spleen cellswere fused with
mouse mveloma cells SP2/0.Monoclonal antibodies(McAb)secreted by hy-bridomas cell lines
3D5,4B2,4F11.5C7.8E3 were screened by EIJISA and cloned.In IFAand IPA staining assay,McAb
4B2 and 8E3 showed strong positive reaction with P. vivaxblood stage parasites,The
cytopla2m of the parasites showed bright green fluorescencewhereas the nucleus did not.The
parasites were clearly visible as dark brown bodies afterimmunoperoxidase staining. McAb
4B2 and 8E3 did not show cross-reaction with the erythrocytic stages of P.
yoelii.
