贮存血红细胞的Ca^(2+)-ATP酶活性和氧耗率观测

摘要 测定24例用ACD保养液于4℃下不同贮存期全血红细胞的Ca2+ATP酶活性。结果表明,贮存2周内酶活性无明显变化,3周后开始下降,至第4周酶活性完全消失。红细胞氧耗量检测结果显示,CPD组的保存效果似较ACD组为佳。提示红细胞膜Ca2+-ATP酶活性可考虑作为血库对贮存血质控的参考指标之一;而氧耗率则有助于对保存效果的检查。 In this study,blood samples were collected from 24 healthy donors and their wholeblood were stored in acid-citrate-dextrose(ACD) anticoagulant separately at 4℃. The mem-brane Ca2+ ATPase activities in the stored RBC were measured following 7,14,21 and 28days after the storage,Whole blood from 3 donors were also selected and divided into twoportions which have been stored in citrate-phosphate-dextrose(CPD) and ACD anticoagu-lants respectively,with the oxygen consumption rate of erythrocytes determined within 10,20 and 35 days after the storage.The results showed that Ca2+ ATPase activitv did notchange obviously until 21 day after the storage and disappeared completely on the 28th day。From the evaluation of oxygen consumption rate, it was found that the efficiency of bloodpreservation in CPD group seemed better than ACD group. Our data suggest that membraneCa2+ ATPase activity may be considered as one of the qualitative criteria for blood preservation and Oxygen consumption rate as an auxillary criterion.
