反相高效液相色谱法测定人血浆及淋巴组织中5─氟尿嘧啶浓度 被引量:3

摘要 本文报道用反相高效液相色谱法测定人血浆及淋巴结内抗癌药物5─氟尿嘧啶的药物浓度。采用UltrasphereODS(250mm×4.6mm,5μm)色谱柱,0.05mol/LNaH2PO4(PH7.1)/CH3OH/H2O(0.5/6/93.5)为流动相,洗脱经乙酸乙酯提取的样品,在254nm处检测药物的峰高,以外标法定量,并对萃取回收率、准确度和精密度等各项指标进行了评价。本文对用5─氟尿嘧啶进行淋巴道化疗的妇癌病人的盆腔和腹主动脉旁各组淋巴结内抗癌药物的浓度进行了监测,获得相应的药代动力学资料,国内尚未见报道。本方法简便、快捷、准确可靠,对临床动态监测5─氟尿嘧啶药浓,指导合理用药具有积极意义。 The method of RPHPLC was reported for determining the concentration of 5-fluorouracil (5-Fu) in plasma and lymph node of gynecological cancer patients. The assay consists of a single extraction of 5-Fu from plasma and lymph node into ethyl acetate and evaporation of organic layer. The residue is reconstituted in mobile phase and injected onto Ultrasphere ODS(250×4.6mm, 5μm) Column, and monitored at 254nm.The mobile phase is a mixture of 0. 05mol/L phosphate buffer(pH7. 1)/methanol/distilled water (0.5/6/93.5). The limit of quantitation for 5-Fu is 0.098mg/L. The extraction recoveries are 73-90%. Winthin-day CVs are 4.79-4.89%. Day to Day CVs are 1.79-4.39%.18 cases were randomly injected with 500mg of 5-Fu for chemotherapy against lymphatic metastases of gynecological cancer via pelvic retroperitoneal cannulation unilaterally while the contralateral side served as control. The determining results shew that the 5-Fu concentration in the lymph nodes on injected sides were 2 to 10 times of that on control sides which was statistically significant (p<0.06).The AUC value on injected sides was times of that on control sides, and the high 5-Fu concentration persisted along period with T 1/2β=21.536 hours. The 5-Fu concentions in the periaorticlymph nodes were relatively high,while the plasma 5-Fu concentrations were low and eliminated rapidly (T1/2β=0. 217 hours).
