内皮素灌流对离体大鼠主动脉c－myc和热休克蛋白70mRNA基因表达的增强作用

The Increase of c-myc and hsp70 Gene Expression in Rat Aortae Induced by Endothelin Perfusion in Vitro

本实验用含内皮素的ＫＨ氏液灌流大鼠离体主动脉６０ｍｉｎ后，提取大鼠主动脉中总ＲＮＡ，用Ｎｏｒｔｈｅｒｎ印迹杂交方法检测ｃ－ｍｙｃ和热休克蛋白７０ｍＲＮＡ转录，放射自显影结果显示：内皮素可诱导大鼠主动脉细胞ｃ－ｍｙｃ和热休克蛋白７０基因表达增加，这在高血压和动脉粥样硬化发病过程中对血管平滑肌的增殖可能起重要作用。

n the study rat aortae were perfused by endothelin in vitro and then RNA sample was ex-tracted from them for Northern blotting in which c-myc and hsp70 gene probes were labeled byα-￣32P-dCTP. The results on the bands of autoradiographs shoW that c-myc and hsp70 gene ex-pression increases in the rat aortae. It might play an important role on proliferation of vascularsmooth muscle cells in hypertension and atherosclerosis.
