摘要
为了探索激光辐射精子的诱变效应,本试验采用TST(TripleStainTechnique,即精子三重染色法)、SPA(SpermPenetraitionAssay,即精子穿透分析法)和CAS(ChromosomalAneu-ploidyinSpermatozoa,即精子单倍染色体制备技术)三项有关精子的现代分析技术,对激光辐射精子的受精学、遗传学效应进行了研究。结果表明:He-Ne激光辐射奶牛冷冻精液,可提高精子存活率,促进精子体外获能、发生顶体反应和芽透卵黄膜;具有明显加速雄性原核形成和诱导精子染色体发生畸变的作用。在本试验条件下,经多次预试验确定的辐射时间中,各指标综合效应最佳剂量组为4分组。该组精子顶体反应率、活精子获能率、卵黄膜穿透率、含原核卵子百分率和染色体畸变率比对照组分别增加了9.45%(P<0.01)、6.89(P<0.01)、12.47%(P>0.05)、17.64%(P<0.05)和46.03%(P<0.05)。
In order to study the effects of He-Ne Laser Irradiation on Holstein bulls spermfertilizalogy and genetics,frozen semen samples five Holstein bulls were tested by using TST(Triple stain Technique),SPA(Sperm Penetration Assay), CAS(Chromosomal Aneuploidyin Spermatozoa).Our results indicated that He-Ne Laser Irradiation is able to increase therate of survival spermatozoa,and to promote spermatozoa in-vitro capaciation,acrosome re-action, percentage of vitelli penetrated,espesially the development of male pronuclei and fre-quency of spermatozoa chromosomal abnormalities;The rate of true acrosome reaction,sur-vival spermatozoa capaciation,vitelli with male pronuclei and spermatozoa chromosomal ab-normalities of 4 minutes group have increased by 9.45%、6.89%、12.47%、17.64%and 46.03%over the control group respectively.
