利用鳞翅目来源的转座子piggyBac建立高效稳定的转基因家蚕技术 被引量:13

摘要 piggyBac转座子元件被成功应用于家蚕转基因.EGFP荧光蛋白基因作为筛选标志,受含有Pax-6结合位点的启动子控制,并能在家蚕的眼部组织中特异性表达.荧光筛选质粒和piggyBac转座子表达质粒被以混合物的形式显微注射到家蚕受精卵,获得出生的G0代蛾子700个,互相交配后,利用EGFP荧光筛选获得111个独立的转基因系.大部分转基因系在基因组上携有2个或多个插入拷贝,并且在整个项目过程中转基因的表型稳定遗传6代以上.对插入位点的序列鉴定发现,其插入位点两侧具有特异性TTAA序列,证实转基因的插入受到piggyBac转座子的调控.高效稳定的家蚕转基因技术的建立为家蚕后基因组的研究和蚕业工业的发展提供了基础.
