摘要 对杂交榛子(代号80-11)进行了离体培养研究.试验结果表明,外植体的选取是离体培养的关键,最佳外植体为:休眠后的基生枝条在室内用营养液培养后萌发的嫩梢.通过最佳培养基组合试验,萌发率可达90%以上,增长系数可达2.2±0.5.诱导阶段的培养基为DKW+BA5mg/L+IBA0.01mg/L+GA30.2~0.3mg/L+葡萄糖30g/L+琼脂6g/L.继代培养时调整BA为3~4mg/L、CGA30.1mg/L.根据在继代过程中出现的内生菌污染问题,试验采用在培养基中加500倍的百菌清来解决. In the present experiment, hybrid hazel (No. 80- 11 ) was employed as the experimental materials. It was found that the explants were the most important factor in vitro propagation. Using newly developed shoots from the forced outgrowth of axillary buds on branches after dormancy as a source of explants could facilitate the establishment of the asepsis system. Germinating rate could reach above 90% and proliferation coefficient could be 2.2 ± 0.5 after culturing on the best medium. At the culture establishment stage, the best medium is DKW + BA5 mg/L + IBA0.01 mg/L + GA30.2 - 0.3 mg/L + glucose30 g/L + agar 6g/L. The concentration of BA was adjusted to 3 ~ 4 mg/L and GA3 to 0. 1mg/L at later subcultures. Internal bacterial contamination could be eliminated in some degree by adding Chlorothalonil to medium.
