肺虚痰阻大鼠肺组织核因子-κB和环氧合酶-2mRNA的表达 被引量:7

摘要 目的:探讨核因子 κB(nuclearfactor kappaB,NF κB)和环氧合酶 2(cycloxygenase 2,COX 2)在 肺虚痰阻大鼠发病中的意义和化痰1号作用的机制。方法:采用二氧化硫烟薰和寒冷刺激建立SD大鼠肺 虚痰阻模型,应用免疫组化法检测正常组、肺虚痰阻模型组及化痰1号治疗组大鼠支气管上皮细胞NF κB 的表达及RT PCR检测肺组织COX 2mRNA的表达。结果:模型组NF κB和COX 2mRNA的表达高于 正常组(P<0.01),中药治疗后,NF κB和COX 2mRNA的表达明显降低,与模型组比较有显著性差异 (P<0.01)。结论:NF κB和COX 2mRNA的表达升高在痰邪致病中起着重要作用,化痰1号可能通过调节 NF κB和COX 2mRNA的表达而起到化痰作用。 Objective: To study the role of nuclear factor-kappa B (NF-KB) and cycloxygenase-2 (COX-2)in the onset of phlegm obstruction due to lung-deficiency in rats and the therapeutic mechanism of Huatan Recipe. Methods. Twenty-four SD rats were randomly divided into normal group, model group and treatment group, with 8 rats in each group. The rats in the model group and treatment group were exposed to sulfur dioxide and cold wind to establish the rat model of phlegm obstruction due to lung-deficiency, and the rats in the treatment group were also treated with Huatan Recipe, a compound traditional Chinese medicine. The expression of NF-KB in the bronchial epithelial cells of the rats was tested with the method of immunohisto-chemistry, and the COX-2 mRNA in the lung tissues of the rats was measured by using reverse transcription-polymerase chain reaction. Results: The expressions of NF-KB and COX-2 mRNA in rats of the model group were higher than those of the normal group ( P〈0. 01 ), and the expressions of NF-κB and COX-2 mRNA in rats of the treatment group were obviously lower than those of the model group (P〈0.01). Conclusion. The NF-KB and COX-2 play an important role in the onset of phlegm obstruction in rats. Huatan Recipe may prevent the development of phlegm obstruction by down-regulating the expressions of NF-κB and COX-2 mRNA.
