摘要 目的通过观察肿瘤坏死因子(TNF)α对肝星状细胞(HSC)形态和结缔组织生长因子(CTGF)表达的影响,探讨TNFα在肝纤维化中的作用机制。方法采用体外培养大鼠肝星状细胞,加入TNFα和TGFβ1,透射电镜观察肝星状细胞的形态学改变并应用逆转录聚合酶链反应(RTPCR)技术检测不同处理组HSC中CTGF的表达。结果TNFα、TGFβ1均诱导HSC中CTGF表达;10μg/L浓度的TNFα作用6、24和48h后,检测到CTGFmRNA表达,而TGFβ1在1μg/L浓度作用4h后,即可诱导HSC中CTGFmRNA表达。结论TNFα诱导HSC中CTGF的表达可能参与早期肝纤维化的形成。 Objective To observe the effect of tumor necrosis factor-α (TNF-α) on morphology and the expression of connective tissue growth factor (CTGF) in hepatic steUate cells (HSC), and explore the mechanism. Methods HSCs were cultured in vitro and treated with TNF-α or TGF-β1. The morphological changes of HSCs were observed and the expression of CTGF in HSC was assessed using reverse transcription-polymerase chain reaction (RT-PCR). Results Both TNF-α and TGF-β1 could stimulate the expression of CTGF mRNA in HSC. The expression of CTGF was detectable in HSC after treatment with TNF-α (10μg/L) for 6, 24 and 48 h. TGF-β1 could induce the CTGF expression just after 4 h of treatment at 1 μg/L concentration. Conclusion TNF-a-induced the CTGF expression in HSC may be the mechanisms involving in the liver fibrosis.
