复力康合剂影响成年雄性大鼠的运动能力 被引量:4

Effects of Fulikang mixture on the exercise ability of adult male rats

摘要 目的:观察复力康合剂提高雄性大鼠运动能力与延缓运动性疲劳的作用。方法:实验于2002-10/2003-02在成都体育学院动物实验室完成。将成年雄性SD大鼠60只随机分为4组,每组15只。空白对照组组仅以蒸馏水灌喂干预;单纯运动组灌喂蒸馏水和负重游泳;低剂量运动及高剂量运动组分别灌喂1.08g/mL,3.13g/mL复力康合剂和负重游泳。游泳训练1次/d,6次/周,4周后处死动物,测定血清睾丸酮、皮质醇、心肌肌浆和线粒体超氧化物歧化酶、谷胱甘肽还原酶活性、丙二醛含量。结果:实验过程中空白对照组,单纯运动组,低剂量运动组,高剂量运动组由于游泳力竭分别死亡6,6,3,8只,最终进入结果分析大鼠37只。①血清睾酮和皮质醇含量:单纯运动组血睾酮显著低于空白对照组犤(418±614)ng/L,(1138±1206)ng/L,(t=2.317,P<0.05)犦,低剂量运动组和高剂量运动组血睾酮显著高于单纯运动组犤(596±456)ng/L,(2565±1307)ng/L,(418±614)ng/L,t=2.296,P<0.05;t=4.672,P<0.01犦;单纯运动组、低剂量运动组、高剂量运动组皮质醇显著高于空白对照组犤(31.4±9.5)μg/L,(31.2±10.1)μg/L,(35.0±7.6)μg/L,(12.2±3.9)μg/L,t=2.612~3.721,P<0.01犦,3组间皮质醇含量基本一致(P>0.05)。②肌浆和线粒体内超氧化物歧化酶、谷胱甘肽还原酶活性和丙二醛含量:单纯运动组和低剂量运动组肌浆和线粒体超氧化物歧化酶活性均显著低于空白对照组犤肌浆:(132.3±15.0)NU/mL,(111.6±11.7)NU/mL,(162.2±17.9)NU/mL;线粒体:(118.1±9.5)NU/mL,(117.8±11.0)NU/mL,(178.3±7.9)NU/mL,t=2.634~3.896,P<0.01犦;单纯运动组和低剂量运动组超氧化物歧化酶活性基本一致。低剂量运动组的线粒体谷胱甘肽还原酶活性显著高于空白对照组犤(1.074±0.243)μkat/L,(0.810±0.242)μkat/L,t=2.447,P<0.05犦,其他各组谷胱甘肽还原酶活性基本一致;低剂量运动组线粒体丙二醛含量显著低于单纯运动组犤(2.92±0.28)μmol/L,(5.14±1.11)μmol/L,t=4.010,P<0.01犦。结论:复力康合剂能有效地改善雄性大鼠的运动性低血睾现象,且高剂量运动组效果更为明显。复力康合剂能降低雄性大鼠心肌线粒体丙二醛含量,提示具有抗脂质过氧化作用。复力康合剂对雄性大鼠超氧化物歧化酶、谷胱甘肽还原酶活性无明显影响。复力康合剂对增强机体运动能力有良性作用。 AIM: To observe the effects of Fulikang mixture in enbancing the exercise ability and delaying exercise-induced fatigue in male rats.METHODS: The experiment was carried out in animal laboratory of Chengdu Institute of Physical Education between October 2002 and February 2003. Sixty adult male SD rats were randomly divided into 4 groups with 15 rats in each group. Rats in the blank control group were only treated with perfusion of distilled water; Rats in the simple exercise group were perfused with distilled water and trained with loaded swimming; Rats in the low and high dose exercise groups were perfused with 1.08 and 3.13 g/mL Fulikang mixture respectively and trained with loaded swimming. The swimming was performed once a day and 6 times every week. All the rats were killed after 4 weeks, the serum levels of testosterene and cortisone, activities of superexide dismutase and glutathione reductases, and content of malondialdehyde in cytoplasm and mitoehondrion of myocardium were determined.RESULTS: During the experiment, 6, 6, 3,and 8 rats died from exhausted swimming in the blank control group, simple exercise group, low and high dose exercise groups respectively, and 37 rats were finally involved in the analysis of results. ① The serum levels of testosterone and cortisone: The serum level of testosterone was significantly lower in the simple exercise group than in the blank control group [(418±614), (1 138±1 206) ng/L,t=2,317, P 〈 0.05], but significantly higher in the low and high dose exercise groups than in the simple exercise group [(596±456), (2 565±1 307),(418±614) ng/L, t=2.296, P 〈 0.05; t=4.672, P 〈 0.01]; The level of cortisone was significantly higher in the simple exercise group and low and high dose exercise groups than in the blank control group [(31.4±9.5),(31.2±10.1), (35.0±7.6), (12.2±3.9)μg/L, t=2.612 to 3.721, P 〈 0.01], and there were no obvious differences among the first 3 groups (P 〉 0.05). ②The activities of superoxide dismutase and glutathione reductases, and the content of malondialdehyde in cytoplasm and mitochondrion: The activities of superoxide dismutase in cytoplasm and mitochondrion were significantly lower in the simple exercise group and low dose exercise groups than in the blank control group [cytoplasm: (132.3±15.0), (111.6±11.7), (162.2±17.9)NU/mL; mitochondrion: (118.1±9.5), (117.8± 11.0), (178.3±7.9) NU/mL,t=2.634 to 3.896, P 〈 0.01], and those were almost the same in the simple exercise group and the low dose exercise group; The activity of glutathione reductases in mitochondrion was significantly higher in the low dose exercise group than in the blank control group [(1.074±0.243),(0.810±0.242) μkat/L, t=2.447, P 〈 0.05], and there were no obvious difference among other groups; The content of malondialdehyde in mitochondrion was significantly lower in the low dose exercise group than in the simple exercise group [(2.92±0.28), (5.14±1.11) μmol/L, t=4.010, P 〈 0.01].CONCLUSlON:Fulikang mixture can effectively improve the lower serum level of testosterone caused by exercise in male rats, and the effect is more obvious in the high dose exercise group. Fulikang mixture can decrease the content of malondialdehyde in mitochondrion of myocardium in male rats,indicating that it has the effect against lipid peroxidation. Fulikang mixture has no obvious influence on the activities of superoxide dismutase and glutathione reductases in male rats. Fulikang mixture plays a good role in enhancing the exercise ability of body.
