MMP-9和TIMP-1在食管鳞癌中的表达及其与淋巴结转移的关系

Expression of MMP-9 and TIMP-1 and Its Correlation with Lymph Node Metastasis in Esophageal Squamous Carcinoma

目的探讨基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶组织抑制因子-1(TIMP-1)在食管鳞癌中的表达及其与淋巴结转移的关系.方法应用免疫组织化学链霉菌抗生物素蛋白-过氧化酶连接法(S-P法)对78例食管鳞癌组织中MMP-9、TIMP-1蛋白表达进行检测.结果 MMP-9在高分化组(Ⅰ～Ⅱ级)及临床早期(Ⅰ～Ⅱ期)病例食管癌的表达明显低于低分化组(Ⅲ～Ⅳ级)及临床晚期(Ⅲ～Ⅳ期)病例的表达(P＜0.05).而TIMP-1在高分化组(Ⅰ～Ⅱ级)及临床早期(Ⅰ～Ⅱ期)病例食管癌的表达明显高于低分化组(Ⅲ～Ⅳ级)及临床晚期(Ⅲ～Ⅳ期)病例的表达(P＜0.05).MMP-9在淋巴结转移组的强阳性表达率明显高于非淋巴结转移组(P＜0.05),而TIMP-1淋巴结转移组强阳性表达率则低于非淋巴结转移组(P＜0.05).MMP-9的表达与食管鳞癌的病理分级和淋巴结转移呈正相关(P＜0.05),而TIMP-1的表达则相反,MMP-9和TIMP-1的表达呈负相关(P＜0.01).结论 MMP-9可作为判断食管鳞癌恶性生物学行为的标记物.MMP-9与TIMP-1与食管鳞癌的浸润和淋巴结转移有关,二者平衡的破坏是食管鳞癌侵袭与转移的重要因素.

Objective To study the expression of matrix metalloproteinase - 9（ MMP - 9） and tissue inhibitor of metalloproteinase - 1 （TIMP- 1）, and its correlation with lymph node metastasis in esophageal squamous earcinoma（KSC）. Methods The expressions of MMP- 9 and TIMP- 1 were detected in 78 eases of esophageal squamous carcinomas by using irmnunohistochemieal SP method. Results The expression level of MMP- 9 in high differentiated （grade Ⅰ-Ⅱ ） and early stage （stage Ⅰ-Ⅱ） ESC was significantly lower than that in low different/ated（grade Ⅲ-Ⅳ ）and late stage （stage Ⅲ-Ⅳ） P 〈0.05）. However, the expression level of TIMP- 1 in high differentiated and early stage ESC was significandy higher than that in low differentiated and late stage ESC （ P 〈0.05）. The expression level of MMP- 9 in ESC was positively correlated with histological grading and lymph node metastasis（ P 〈 0.05）, but the expression level of TIMP - 1 in ESC was negatively correlated with those（ P 〈0.05）. There was a negative correlation between MMP - 9 and TIMP - 1 expression（ P 〈 0.01）. Conclusion MMP - 9 might be a biomarker for evaluating the malignant biological behavior of ESC, and the expression levels of both MMP - 9 and TIMP - 1 were related with the invasion and lymph node metastasis of ESC.
