摘要
目的应用60Coγ射线照射、液氮深低温保存大鼠同种带瓣肺主动脉(VPHC),并建立VPHC移植模型,观察不同吸收剂量对同种带瓣管道免疫原性的作用,寻找合适吸收剂量,为进一步基础研究及临床应用提供依据。方法应用流式细胞仪(FCM)检测CD4、CD8细胞亚群细胞,凋亡检测用TUNEL法,ICAM-1用免疫组化检测。结果γ射线照射有效地抑制了VPHC移植术后CD4+、CD8+和ICAM-1的表达量,减轻了排斥反应的发生,但同时也导致细胞凋亡甚至发生细胞坏死改变,降低了VPHC的活性,影响其耐久性。结论600Gy60Coγ射线照射在不严重损伤大鼠VPHC活性的同时可明显地降低VPHC移植术后免疫排斥反应的发生,可作为参考剂量用于降低VPHC免疫原性的研究。
Objective To observe the effect of ^60Co γ on immunogenicity of valved pulmonary homograft conduits(VPHC) in order to determine the proper radiation dose. Methods CD4 and CD8 subset was detected by FCM, apoptosis was measured by TUNEL and ICAM-1 was detected. Results ^60Coγ/effectively inhibit CD4^+, CD8^+ and ICAM-1 expression, cause could apoptosis or even cellular necrosis at the same time and depress the VPHC activity. Conclusions Rejection can be decreased remarkably without damaging VPHC activity of rats when the ^60Coγ dose is below 600Gy, it can be a reference dose for the study of VPHC immunogenicity surpressed by 60Co γ.
