摘要
目的探讨奥曲肽对肝癌微血管密度(MVD)和血管内皮生长因子(VEGF)表达的影响。方法分别于16只裸小鼠右腋窝皮下注射HepG2细胞建立动物模型后,随机分为2组,每组各8只。实验组:皮下接种肿瘤1d后给予奥曲肽[100μg/(kg·d)]腹腔注射,连用14d;对照组:予以相同容积的无菌生理盐水腹腔注射,连用14d。14d后处死小白鼠,检测肿瘤大小、血管密度(MVD)及血管内皮生长因子(VEGF)。结果皮下接种7d后实验组肿瘤的体积均明显小于对照组(<0.01)。实验组MVD、VEGF表达明显低于对照组(P分别<0.01和0.05)。结论奥曲肽对裸小鼠肝癌移植瘤具有抑制作用,对肿瘤MVD及VEGF的抑制作用可能为其机制之一。
[Objective] To evaluate the effect of 0ctreotid on the expression of MVD andVEGF in seeding carcinoma. [Meghods] To inject 0.2 mL HepG2 cells into subcutaneous tissue at right axilla for preparation animal mode respectively in 16 nude mice,and then randomized into two groups; Experimental and control group, 8 mice each group. Experimental group: Every one received octreotide (100 μg/(kg·d)×14 days) intraperitoneal injection after implantation, control group: each mouse received same amount of saline solution intraperitoneal injection. Subcutaneous tumor' s size was measured everyday. All mice were killed on 14d after implantation, and the tumors werere moved for immunohistochemical exantination of MVD AND VEGF. [Results] Mean volum of tumors was smaller in experimental group than in control group after 7 days (0.01). The expression of MVD and VEGF was significantly lower in experimental group than in control group (P 〈0.01 and 0.05 respectively). [Conclusion] Octreotid can inhibit seeding tumor of HepG2 cells in nude mice. Inhibition of tumor angiogenesis maybeone of the mechanisms of its antltumor effect by inhibition of MVD andVEGF.
