急性心肌损伤细胞因子变化和N-乙酰半胱氨酸的干预作用 被引量:1

摘要 目的研究N-乙酰半胱氨酸(NAC)对异丙肾上腺素(ISO)致大鼠急性心肌损伤的保护作用及作用机制。方法用异丙肾上腺素建立心肌缺血坏死模型,30只雄性Wistar大鼠,随机分为3组:ISO组、ISO+NAC组、对照组。测定大鼠血清心肌酶CK、LDH含量,测定大鼠血清和心肌组织脂质过氧化物丙二醛(MDA)含量,RT-PCR测定心肌组织中细胞因子TNF-α、IL-1β、INF-γmRNA的表达。结果与ISO相比较,ISO+NAC组大鼠血清CK、LDH含量显著降低,大鼠血清和心肌组织MDA含量显著降低;心肌组织中细胞因子TNF-α、IL-1β、INF-γmRNA表达显著降低。结论NAC对ISO致大鼠心肌损伤有保护作用,其机制可能与减少心肌过氧化损伤,减少细胞因子基因的表达有关。 [Objective] The present study was undertaken to study the protective effects of N-acetyl-L-cysteine on acute heart injury induced by isoproterenol in rats and its mechanisms. [Methods] Thirty male Wistar rats were randomly divided into three groups: ISO, ISO+NAC and control group. Serum content of CK and LDH were examined; Serum and myocardium content of MDA were examined; myocardium TNF-α、IL-1β、INF-γgene expression were examined by RT-PCR analysis. [Results] Compared with ISO group, serum content of CK and LDH were decreased significantly in ISO+NAC group; Serum and myocardium content of MDA were decreased significantly in ISO+NAC group; meanwhile myocardium expressions of TNF-α、IL-1β、INF-γdescreased significantly in ISO+NAC group. [Conclusion] NAC has preventive effects on acute heart injury induced by isoproterenol in rats, which is probably associated with decreasing reactive oxygen species and reducing cytokines gene expression.
