MGMT基因在脑胶质瘤治疗中的意义 被引量:2

摘要 胶质瘤是颅内肿瘤中发病率最高的肿瘤,目前临床上多采取以手术治疗为主辅以化学治疗和放射治疗.氯乙基亚硝脲类烷化剂(CNUs),以其高脂溶性和易于透过血脑屏障的特性,长期以来被看为治疗脑胶质的最有效的化疗药物.但抗药性的产生使其临床有效率不足30%,而成为脑胶质瘤化疗失败的主要原因.06-甲基鸟嘌呤-DNA甲基转移酶(MGMT)修复亚硝类药物所造成的DNA损伤,是肿瘤细胞对亚硝类耐药的主要分子机制[1].有研究表明,73%的脑肿瘤有MGMT的表达[2];且MGMT活性与抗药性呈正相关,降低细胞中MGMT的活性将会逆转细胞的耐药表型,增强亚硝类药物的抗肿瘤效果[3].因此,探索抑制细胞内的06-甲基鸟嘌呤DNA甲基转移酶(MGMT)基因表达水平的有效方法,对于提高脑胶质瘤的化疗效果具有重要的临床意义.
