超声辅助转基因治疗缺血性心脏病的骨髓间充质干细胞的实验研究 被引量:1

摘要 目的血管内皮生长因子(VEGF)基因转染骨髓间充质干细胞,探讨骨髓间充质干细胞定向分化血管内皮细胞的可行性研究。方法VEGF165基因的质粒PEGFP-C1,采用超声辅助电穿法转染骨髓间充质干细胞,转染后于荧光显微镜下观察绿色荧光蛋白(GFP)表达情况。结果超声辅助转染后5h可见细胞内有GFP表达,10h后出现表达高锋,持续3d左右,其后有些细胞荧光开始减退。结论VEGF基因转染骨髓间充质干细胞后,骨髓间充质干细胞内有GFP表达,提示细胞转染成功,骨髓间充质干细胞定向分化为血管内皮细胞具有可行性。 Objective Using vascular endothelial growth factor (VEGF) genes to transfect medullary mesenchymal stem cells,then to investigate the feasibility of medullary mesenchymal stem ceils directionally differentiating into vascular endotheliocytes. Methods Using VEGF165 genes plasmid PEGFP-C1 was transfected into medullary mesenchymal stem ceils by ultrasonic-assisted electrical penetration, then the expression of green fluorescence protein(GFP) was observed under the fluorescence microscope. Results GFP expression in the ceils could be found 5h after ultrasonic-assisted transfection, and which reached to peak in 10h, persisted for 3 days, then some of fluorescence in the ceils began to disappear. Conclusion After VGEF genes transfecting medullary mesenchymal stem ceils, there is GFP expression in the medullary mesenchymal stem ceils, which suggests that the transfection is successful and it is feasible for marrow medullary mesenchymal stem ceils directionally differentiating into vascular endotheliocytes.
