摘要
目的探讨大鼠胃缺血-再灌注损伤时胃窦黏膜D细胞的形态变化。方法SD雄性大鼠随机分为正常对照组(CON)、单纯缺血组(ISC)、缺血-再灌注1h组(IR1)、6h组(IR2)和24h组(IR3),用免疫组织化学方法观察大鼠胃缺血-再灌注损伤过程中胃窦黏膜D细胞的形态变化。结果正常组大鼠胃窦黏膜中D细胞主要分布于幽门腺的中、下13处,形态多样;和正常组比较,D细胞的数目在ISC组和IR1组明显减少,细胞多为圆形,很少见有胞质突起;在IR2组和IR3组则可见D细胞的数目明显增多,形态多样,并可见D细胞也分布于幽门腺上13处。结论胃窦黏膜中D细胞的形态数量在大鼠胃缺血-再灌注过程中有变化,提示生长抑素参与了胃缺血-再灌注损伤过程。
Objeetive To investigate the morphological changes of gastric ischemia - reperfusion injury (GI - RI) of D cells in the mt antral gastric mucesa. Methods Sprague - Dawley male rots were randomly divided into control (CON), ischemia (ISC), ischemiarepeffusion (I/R) groups. The I/R group was further divided into I/R1, I/R2 and I/R3 groups to undergo reperfusion for 1 h, 6 h and 24 h respectively. The animals were killed 1,6,24 hours afer reperfusion separately in each group. The morphological changes of GI - RI of D cells in the rat antral gastric mucosa were studied by using immtmohistochemical method. Results The D cells were polymorphic in shape, predominantly located in the middle lower zone of the pyloric gland in the normal control group. In the ISC and I/R1 groups, the D cells were decreased in number, became rotmd in shape, with few protrusion on surface. In I/R2 and I/R3 groups, the D cells were markedly increased in number, varied in shape, and were also noticed in the upper zone of the pyloric glands. Conclusion The findings indicate that there are alterations in morphology and number of D cells in rat antral gastric mucosa in response to isehemia- repeffusion, suggesting somatostatin be involved in the process of gastric ischemia - reperfusion injury.
