薯蓣皂苷元对人胃低分化粘液腺癌细胞作用的研究 被引量:11

摘要 目的观察薯蓣皂苷元(Diosgenin,Dio)对人胃低分化粘液腺癌(MGC-803)细胞的作用,并初步探讨其作用机制。方法MTT法测Dio对MGC-803细胞的抑制作用;考察Dio对MGC-803细胞分裂和集落形成能力的影响;采用3H-TdR掺入法检测Dio对MGC-803细胞DNA合成的影响。结果一定浓度范围内Dio对MGC-803细胞的增殖有剂量、时间依赖性抑制作用;Dio明显抑制MGC-803细胞分裂和集落形成,半数抑制浓度(IC50)为13.17mg/L,Dio能显著抑制MGC-803细胞DNA合成。结论Dio对MGC-803细胞的抑制作用与抑制该细胞DNA合成有关。 Objective To observe the effect of diosgenin (Dio) on poor differentiated adenocarcinoma (MGC-803) cells of human stomach and discuss the mechanism. Methods MTr assay was used to determine the inhibitory effect of Dio on MGC-803 cells, and to observe the influence of Dio on the ability of cell cleavage and colony formation. The method of 3^H-TdR was used to detect the influence of Dio on DNA synthesis of MGC-803 cells. Result It was found that Dio inhibited the growth of MGC-803 ceils in a dose-time dependent manner at a certain range of concentration, and inhibited obviously the cell cleavage and colony formation. IC50 was 13.17 μg/mL. Dio inhibited obviously DNA synthesis of MGC-803 cells. Conclusion The inhibitory effect of Dio on MGC-803 cells is related to its inhibitory effect on DNA synthesis.
