摘要
目的观察没食子酸丙酯对大鼠缺血再灌注心肌线粒体功能的影响作用。方法结扎大鼠冠状动脉30 min再灌注20 min,测定心肌线粒体中琥珀酸脱氢酶(SDH)、细胞色素氧化酶(CCO)、Ca2+.Mg2+—ATP酶、Ca2+—ATP酶、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶((GSH—PX)活性及细胞色素、磷脂和丙二醛含量。结果没食子酸丙酯(20和40 mg/kg,结扎冠状动脉前60 min ip)能显著减轻缺血再灌注所致琥珀酸脱氢酶(SDH)、细胞色素氧化酶(CCO)、Ca2+.Mg2+-ATP酶、Ca2+—ATP酶、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶活性的抑制(P<0.05)及细胞色素aa3、细胞色素C和膜磷脂含量的减少,并能抑制丙二醛含量的升高。结论没食子酸丙酯对缺血再灌注时心肌线粒体功能具有保护作用,其机制可能是通过提高对氧自由基的清除功能和抑制脂质过氧化。
Objective To study the effect of propyl gallate on myocardial mitochondria induced by ischemia/reperfusion in rats. Methods Ischemia/reperfusion injury was induced by coronary artery ligation for 30min and reperfusion for 20rain. The activity of Ca^2+- ATPase, Ca^2+·Mg^2+- ATPase, succinct dehydrogenase(SDH), cytochrome oxidase(CCO), SOD, GSH- PX, and the content of Cytaa3 and Cyt C, phospholipid (PL) and MDA in the myocardial mitochondria were measured. Results Treatment with propyl gallate significantly reduced the decreased activity of Ca^2+- ATPase, Ca^2+·Mg^2+- ATPase, SDH, CCO, SOD and GSH-PX (P 〈 0.05), and the decreased content of Cyt C, Cytaa3 and PL, and attenuated the increased content of MDA induced by ischemia/reperfusion. Conclusion Propyl Gallate has a protective effect on injury of myocardial mitochondria induced by ischemia/reperfusion in rats, and the effects of propyl gallate are related to scavenging oxygen free radicals and reducing lipid proxidation.
