摘要
目的探讨补肾活血方对精索静脉曲张模型大鼠生精细胞凋亡及各级生精细胞数的影响。方法将50只3月龄雄性SD大鼠先随机抽出10只为假手术组,其余40只大鼠采用肾静脉缩窄法建立精索静脉曲张病理模型,造模完成后再随机均分为模型组,补肾活血方低剂量组、中剂量组、高剂量组。造模后48 d开始给药,连续给药60 d,用流式细胞仪检测各组大鼠睾丸生精细胞凋亡数、凋亡率及各级生精细胞数。结果假手术组未出现明显的凋亡峰,模型组与各治疗组均出现明显的凋亡峰,且模型组大鼠睾丸生精细胞凋亡率显著高于各治疗组(P<0.01)。各级生精细胞数比较,1C细胞(精子细胞与精子)数模型组与各治疗组比较有显著性差异(P<0.05);2C细胞(次级精母细胞)数各组间比较无显著性差异;4C细胞数(初级精母细胞)模型组与治疗组比较有非常显著性差异(P<0.01)。结论生精细胞大量凋亡是精索静脉曲张导致不育的重要病理机制,补肾活血方能降低生精细胞凋亡率,提高模型动物各级生精细胞数量。
Objective To study the effects of experimental varicocele on the apoptosis of the rat spermatogenic cells, and the effects of Bu Shen Huo Xue Prescription on it, to investigate the mechanism of Bu Shen Huo Xue Prescription for male sterility with vaficocele. Methods Fifty Spragne-Dawley( S. D) male rats were divided into five groups: control group, model group, Bu Shen Huo Xue Prescription low dose group, middle dose group, high dose group. Experimental varicocele was created in S. D male rats by partial ligation of left renal vein. Forty-eight days after modeling, model rats were fed with medicine for sixty days. The method of flow cytometry was used to measure the apoptosis number spermatogenic cells. Results There was no marked apoptosis peak in control group, in other groups there were marked apoptosis peaks, and the apoptosis peak in model group was significantly higher than those in treatment groups ( P 〈 0.01 ). The numbers of 1C and 4C between model group and treatment groups were significantly different ( P 〈 0.05 and P 〈 0.01 ),and the numbers of 2C between model group and treatment groups were the same. Conclusions Varicocele can induce great apoptosis of spermatogenic cells and the decrease of spermatogenic cells. Bu Shen Huo Xue Prescription can reduce the apoptosis number of spermatogenic cells and increase the number of spermatogenic cells, this may be the mechanism of Bu Shen Huo Xue Prescription for male sterility with varicocele.
