嗅鞘细胞复合细胞外基质凝胶移植对周围神经损伤后运动神经元的保护作用

摘要 目的探索周围神经损伤后嗅鞘细胞(OECS)和细胞外基质(ECM)凝胶复合移植对周围神经运动元的保护作用。方法将Wistar大鼠20只随机分为实验组、对照组。每只大鼠均切断右侧坐骨神经,之后以硅胶管套接于坐骨神经切断部位,实验组囊内给予OECS制成1×106/ml的ECM凝胶,对照组囊内给予磷酸缓冲液(PBS)0.05ml。术后2周,应用酶组织学方法进行相应阶段脊髓前角α运动神经元一氧化氮合酶(NOS)和乙酰胆碱脂酶(AchE)染色,通过标准网格法进行神经元计数并应用计算机图像分析测定胞体的直径和面积。结果与正常侧相比,对照组损伤侧脊髓前角α运动神经元NOS活性明显增高,实验组差异无显著性(P>0.05),损伤侧实验组3.8±1.4,对照组11.4±2.6,差异非常显著性。对照组损伤神经元胞体直径(27.83±5.28)cm,面积(630.24±252.27)cm2,未损伤侧直径(23.31±3.87)cm,面积(438.47±142.14)cm2,损伤明显增加。结论周围神经损伤后,嗅鞘细胞复合细胞外基质凝胶对脊髓前角运动神经元具有保护作用。 Objective To study the protective effect of oLfactory entheathing cells (OECs) cooperation with ECM Gel to motonearons after sciatic nerve injury in rats. Methods Twenty wistar mice were divided into 2 groups randomly,The right sciatic nerve of each mouse was resected.After Silicone-tuhlization capsule of sciatic nerve in rats.OEC,were seeded into 30% ECM at 1×10^6/ml and PBS Solution contral were injected into the capsule respectively, Enzyme histoehemistry was used to determine Ache and NOS activity in spinal a motoneurons in spinal segments 2 weeks later.Nearon were counted, and the diameter and area of the neurons were analyzed by computer image analysis system. Results compared with the un-affected side, the activity of both AchE and NOS was Significantly increased in the motoneurons of the injured side in contral group. The Change was not Significant in the experimental group (P〉0.05).The number of NOS activity was 3.8±1.4 in experimental group and area of neurons on the injured guoup and area of neurons on the injured Side of the control group(27.83±5.28)cm and (630.24±252.27)cm^2 respetively ,significantly greater than those on the un-affeted side(23.31±3.87)cm and (438.47±142.14)cm^2 respectively. Conclusions OECs,cooperation with ECM Gel can protect a motoneurons from degeneration and necrosis after peripheral nerve injury.
