摘要
目的:观察大黄素对急性坏死性胰腺炎(ANP)大鼠核转录因子κB(NFκB)变化的影响,探讨大黄素治疗ANP的作用机制。方法:将60只SD大鼠随机分为假手术组、ANP组、大黄素治疗组;观察各组大鼠血清淀粉酶、肿瘤坏死因子α(TNFα)、白细胞介素6(IL6)含量及胰、肺组织NFκB活化表达。结果:与假手术组比较,ANP组TNFα、IL6含量均明显升高,胰、肺组织NFκB表达亦增加(P均<0.01)。应用大黄素治疗后,其各项指标均显著低于ANP组,胰、肺组织NFκB活化表达降低(P均<0.01),24h大鼠死亡率低于ANP组(P<0.05)。结论:大黄素治疗ANP的作用机制可能是通过抑制NFκB活化、下调细胞因子表达来减轻ANP的炎症反应。
Objective: To investigate the effects and mechanism of emodin on nuclear factor - κB(NF-κB) expression in acute necrosis pancreatitis (ANP) in rats. Methods: Sixty SD rats were randomly divided into sham operation group, ANP group and emodin treatment group. ANP was induced by intraductal administration of 5% sodium taurocholate. Emodin treatment group was given emodin 0.25 mg/100 g by administering intravenously at the time of induction of pancreatitis. Ten rats were killed at 6 hours after induction of the model and contents of serum amylase, interleukin -6 (IL -6), tumor necrosis factor-α(TNF -α) as well as NF -κB in pancreas and lung tissues were detected. Meanwhile the pathology of pancreas and lung tissues was observed. Others were used to observe the mortality rate. Results: Serum amylase,IL - 6, TNF-α as well as NF-κB in pancreas and lung tissues in ANP group all increased compared to thosein the sham operation group (all P^0.01), and they decreased much more in emodin treatment group compared to those in ANP group (all P〈0.01).The mortality rate was lower in emodin treatment group compared to that in ANP group(P〈0.05). Conclusion: Emodin plays a role in the treatment of pancreatitis.It may depress the expression of NF-κB, down-regulate the cytokines and ameliorate the inflammatory response of ANP.
