肾炎大鼠肾组织巨噬细胞与细胞外基质的动态关系

Dynamic relationship between macrophage and extra cellular matrix in the kidney of rats with experimental nephritis

摘要 目的:观察肾毒血清肾炎(NTN)发病过程中肾组织巨噬细胞(Mф)浸润与细胞外基质(ECM)含量的动态变化。方法:用兔抗鼠肾毒血清复制NTN模型。在肾炎不同时间点(3、7、15、30和90d)行肾活检,HE和Mas-son’s染色,光镜观察肾组织形态,免疫组化ABC法检测Mф、纤维连接蛋白(FN)、Ⅲ和Ⅳ型胶原。结果:NTN发病3d,肾组织已有Mф浸润,先于FN和胶原的变化。NTN15d,肾炎动物均有明显蛋白尿,血肌酐升高,Mф明显浸润和ECM沉积。NTN90d,7只动物肾功能和肾组织改变明显减轻,Mф浸润明显减少,另外5只动物肾组织明显纤维化及Mф浸润,并且Mф浸润程度与肾功能和FN、Ⅲ和Ⅳ型胶原沉积呈正相关。结论:肾组织Mф促进ECM沉积而参与NTN进行性恶化或好转的机制。 AIM: To observe the changes in infiltrating maerophages (Mφ)and extraeellular matrix (ECM) in the kidney in the progressive course of nephrotoxic nephritis (NTN). METHODS: NTN model was established with rabbit - anti - rat nephrotoxic serum. On day 3, 7, 15, 30 and 90, renal biopsies were performed. Renal histology was checked under light microscopy with HE and Masson's trichrome staining sections. Mφ, fibronectin (FN), type Ⅲ and Ⅳ collagen were examined with immunohistochemistry ABC method. RESULTS: Infiltration of Mφ appeared on day 3 of NTN and preceded changes of FN and collagen. On day 15 of NTN, all nephritic animals had significant proteinuria, increased serum creatinine, infiltration of Mφ and deposit of entracellular matrix. On day 90 of NTN, seven nephritic animals improved significantly, while other five developed renal scarring with diffuse infiltration of Mφ which positively paralleled to renal function and deposit of FN, type Ⅲ and Ⅳ collagen.CONCLUSION: Mφ infiltrating into renal tissue enhances deposit of ECM and therefore plays important roles in progression or improvement of NTN.
