彗星试验检测叔丁基对苯二酚对苯骨髓毒性的保护作用

Detecting Protective Effect of Tert-butylhydroquinone (tBHQ) on Cytotoxicity of Bone Marrow Cells Induced by Benzene with Comet Assay

摘要 目的 用彗星试验检测叔丁基对苯二酚(tert- butylhydroquinone,t BHQ)对苯诱导的骨髓细胞毒性的保护作用。方法 体外培养的大鼠骨髓细胞行t BHQ(10 0 μm ol/ L)预处理12 h后以不同浓度(0、5、10、15、2 0 mm ol/ L)苯分别染毒2、4、6 h。用彗星试验检测骨髓细胞的DNA损伤,计算DNA迁移率和迁移度。结果 随苯染毒浓度的增加,染毒时间的延长,骨髓细胞DNA迁移率和迁移度增加。t BHQ预处理后5、10、15、2 0 mm ol/ L 苯染毒骨髓细胞的DNA迁移率和迁移度均减低(P<0 .0 5 )。结论 苯可诱导骨髓细胞DNA损伤,并呈现一定的时间、剂量依赖性;叔丁基对苯二酚对苯诱导的骨髓细胞毒性有保护作用。 Objective To investigate the protective effect of tert-butylhydroquinone (tBHQ) on cytotoxicity to bone marrow cells induced by benzene in vitro with comet assay. Methods Rat bone marrow cells were divided into two groups randomly. Cells of control group were stimulated by 0, 5,10,15,20 mmol/L benzene for 2,4,6 h respectively after being treated by 100μmol/L tBHQ for 12 h. DNA damage was assessed with comet array by calculating DNA migration and the lengths of DNA migration of bone marrow cells. Results DNA damage of bone marrow cells increased with the increase of benzene concentration and time of treatment. DNA migration and the lengths of DNA migration of rats bone marrow cells under 5,10,15,20 mmol/L benzene treatment in tBHQ-pretreated group were significant lower than those in control group at the same time point(P〈0. 05). Conclusion It indicates that benzene can induce DNA damage of bone marrow ceils in a time-dependent and dose-dependent manner to some extent, tBHQ can protect bone marrow ceils against cytotoxicity induced by benzene.
