线栓法制备Wistar大鼠局灶性脑缺血模型的实验研究 被引量:21

摘要 目的:建立一种简便可靠,创伤较小的Wistar大鼠局灶性脑缺血模型。方法:参照Zealonga及Koizum的线栓法,加以改进,制作Wistar大鼠局灶性脑缺血模型,随后行神经病学检查,测定脑组织含水量、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性,并进行病理组织学检查,以观察模型的可靠性。结果:大鼠大脑中动脉阻断2小时(MCAO2h)后,出现神经功能障碍,缺血脑组织含水量及MDA含量增加,SOD活性下降,HE染色显示明显脑水肿。结论:该改良法制备的模型简便易行,结果可靠,创伤小,稳定性好,是用于研究局灶性脑缺血较为理想的实验动物模型。 Objective: To establish a simple, reliable and little trauma model of focal cerebral ischemia in wistar rots.Methods: The focal cerebral ischemia model in wistar rats were made by the modified suture - occuluded method of Zcalonga and Koizum. Then the reliability of this model was evaluated with the examination of neurological function, the contents of MDA and water, the activities of SOD, HE staining. Results: 2 hours after MCAO,rats showed neurologincal functional obstacle,contents of MDA and water in brain tissue were increased obviously, actvities of SOD were markedly decreased,HE staining clearly indicated the brain edema. Conclusion:The improved model is simple, reliable, little trauma and stable.It is advisable to use this experimental animal model to study focal cerebral ischemia.
