摘要
目的:研究迷走神经刺激(VNS)对大鼠部分点燃和全面点燃癫痫行为及杏仁核放电的抑制作用的差异。方法:用频率16Hz,波宽1.0ms,串长10s,串隔7min,强度3.0mA的恒流脉冲电刺激15只大鼠杏仁核,观测部分点燃(癫痫部分性发作)大鼠左侧颈迷走神经干7次及32次、全面点燃(癫痫全身性发作)大鼠的左侧颈迷走神经干7次。同时用0.4mA的恒流脉冲电刺激各组大鼠的杏仁核,观察三组动物的发作抑制率(SIR),部分与全面点燃组的平均行为级数下降比(MBSDR),平均后放电时间下降比(MADTDR)和平均后放电阈值上升比(MADTIR)的差异。结果:部分点燃VNS7次组平均SIR为4.30%,部分点燃VNS32次组平均SIR为20.05%,全面点燃VNS7次组平均SIR为66.67%(P<0.01)。部分与全面点燃组的MBSDR分别为0.27±0.16、0.46±0.25,MADTDR分别为-0.05±0.23、0.63±0.32,组间差异非常显著(P<0.01)。两组MADTIR分别为1.86±0.75、1.79±1.08,组间无显著差异(P>0.05)。结论:VNS对大鼠全身性发作的发作频率和发作强度的抑制作用优于对部分性发作的发作频率和发作强度的抑制作用。
Objective: To study quantitatively and comparably the difference of anti-epilepsy effects of vagus nerve stimulation(VNS) between focal kindling and generalized kindling in rats. Methods: The left cervical vagus nerves of 15 focal kindling (partial seizures) rats were stimulated 7 or 32 times, and 15 generalized kindling (generalized seizures) rats were done 7 times with constant square wave pulse current with frequencies of 16 Hz, pulse durations of 1.0 ms, train durations of 10 s, interstimulus interval of 7 min and intensities of 3. 0 mA. The amygdalas were stimulated with the constant current with intensities of 0. 4 mA in the same time. Then the seizure inhibition rates(SIR) were observed in these three groups. The mean behaviour seale(MBS) decrease rate, mean after discharge time (MADT) decrease rate and mean after discharge threshold(MADT) increase rate were observed. Results: The SIR were 4.30% in the partial seizure VNS 7 times group, 20. 05% in the partial seizure VNS 32 time and 66. 67% in the generalized seizure group, respectively (P〈0. 01). MBS decrease rates in the partial seizure VNS 7 times group and generalized seizure group were 0. 27±0. 16 and 0. 46±0. 25(P〈0. 01), respectively, and MADT decrease rates in this two groups were -0. 05±0. 23 and 0. 63±0. 32(P〈0. 01). MADT increase rates in the partial seizure VNS 7 times group and the generalized seizure group were 1. 86±0. 75 and 1. 79±1. 08 (P〉0.05).Conclusion: The inhibitive effects of VNS on seizure frequencies and intensities in the generalized seizure group are more marked than in the partial seizure group.
