摘要
目的:观察普伐他汀对内皮祖细胞(endothelial progenitor cells,EPCs)培养中扩增及黏附能力的影响。方法:采用密度梯度离心法分离人脐血单个核细胞,接种到包被有人纤维连接蛋白的培养皿,分为对照组(未加细胞因子)、V组(加VEGF10 ng/m1)、P0.1组(加普伐他汀0.1 μmol/L)、P0.01组(加普伐他汀0.01μmol/L),培养6d后,采用流式细胞术PE-CD34及FITC-VE-Cadherin双标法测定各组扩增细胞数, 采用黏附能力测定试验测定黏附能力。结果:V组、P0.1组、P0.01组的扩增EPCs数显著高于对照组(P< 0.01),且P0.1组、P0.01组均显著高于V组(P<0.001)。P0.1组、P0.01组的黏附EPCs数显著高于对照组(P<0.001)。结论:普伐他汀可显著提高培养EPCs的扩增能力及黏附能力。
Objective: To investigate the effects of Pravastatin on expand and adhesion ability of endothelial progenitor cells (EPCs). Methods: Mononuclear fraction of human umbilical cord blood was obtained by Ficoll density gradient centrifugation. Isolated ceils were plated on fibronectin coated culture dishes. Ceils were divided into four groups: group control, group Ⅴ, group PO.1, group P0.01(supplemented with none cytokine, VEGF10ng/ml, 0.1μmol/L Pravastatin, 0.01μmol/L Pravastatin. respectively), after six days in culture, fluorescenceactivated cell sorting analysis of PE-CD34 and FITC-VE-Cadherin was performed to detect the numbers of the expand of EPCs in each group, adhesion assay was performed by replating cells on fibronectin coated dishes, and then counting adherent cells. Results: Numbers of expand EPCs of group Ⅴ, group P0.1 and group P0.01 were significantly inceased as compared with those of group control(P〈0.01), and numbers of expand EPCs of group P0.1 and group P0.01 were more than those of group Ⅴ(P〈0.001).Numbers of adhesive EPCs of group P0.1 and group P0.01 were significantly inceased as compared with those of group control(P〈0.001). Conclusion: Pravastatin can significantly enhance the expand and adhesion ability of endothelial progenitor cells.
