摘要 目的:研究霉酚酸(Mycophenolicacid,MPA)诱导T淋巴细胞白血病细胞株Molt-4凋亡的作用及其机制。方法:光学显微镜及透射电镜观察细胞凋亡的形态学特征,琼脂糖凝胶电泳检测DNA断裂,流式细胞仪分析细胞AnnexinV表达、周期变化及caspase-3活性。人Burkitt淋巴瘤细胞株Raji作为对照。结果:MPA诱导Molt-4细胞的凋亡率与作用浓度和时间成正比;细胞被阻滞于S期。MPA作用24h后细胞内caspase-3活性增高,呈浓度依赖性。MPA不诱导Raji细胞凋亡。结论:MPA可诱导Molt-4细胞凋亡,其作用可能与激活caspase-3有关。 Objective: To determine whether mycophenolic acid (MPA) exerts an apoptotic effect on human leukemic T cells Molt 4 and to elucidate its possible mechanism. Methods: Cell morphology,DNA fragmentation, cell cycle,percentage of annexin V positive cells and enzymatic activity of caspase-3 were measured by microscopic, electrophoretic and flow cytometric techniques respectively. Human leukemic B cell line Raji was used as control. Results: MPA could induce apoptosis of Molt 4 cells with dose- and time-dependent manners; cells were blocked in the S phase. The activity of caspase 3 was enhanced in a dose-dependent manner in Molt-4 cells treated with MPA for 24 h. MPA could not induce apoptosis in Raji cells. Conclusion: MPA can induce apoptosis in Molt 4 cells which may be involved in the activation of caspase-3.
