摘要
目的:观察亚硒酸钠对大鼠肾小球系膜细胞(RGMC)表达环氧化酶-2(COX-2)的影响,从而研究硒在抗糖尿病肾病中的作用机制。方法:分别以高葡萄糖、糖基化终产物,高胰岛素和过氧化氢孵育RGMC;先给予亚硒酸钠预处理细胞后,再分别给予上述4种因素孵育RGMC,观察细胞COX-2 mRNA的表达。结果:4种刺激因素均可作为独立因素,导致RGMC COX-2 mRNA表达量的增加;亚硒酸钠能抑制上述4种因素所致的COX-2 mRNA的表达。结论:亚硒酸钠通过抑制COX-2 mRNA在RGMC中的表达,从而有效地防治糖尿病肾病的发生发展。
Objective :To observe the effect of sodlium selenite on expression of Cyclooxygenase - 2 (COX-2 ) mRNA in rat mesangial cells (RGMC), and to study the mechanism of ,sodium selenite in anti diabetic nephropathy. Methods :RGMC was incubated with high glucose, advanced glycosylation end products (AGE), high insulin and H2O2 respectively; simultaneously, RC-MC pre- treated with sodium selenite was incubated with above factors. The expre,ssion of COX- 2 mRNA was detected and compared. Results:The four factors increased the expre,ssion of COX-2 mRNA independently;sodium selenite inhibited the expression of COX-2 mRNA by the four factors distinctly. Conclusion :Sodium selenite suppresses the expression of COX-2 mRNA in RGMC, which suggests that sodium selenite may play a significant role in the prevention of diabetic nephropathy by inhibition of the expression of COX-2 mRNA in RGMC.
