新鲜羊膜与保存羊膜中HGF、bFGF表达的对照研究 被引量:4

摘要 目的:研究肝细胞生长因子(HGF)、碱性成纤维细胞生长因子(bFGF)在新鲜和两种保存羊膜中的表达,探讨新鲜和保存羊膜的生物学活性差异。方法:取健康剖宫产产妇胎盘羊膜,随机分成三组:新鲜组、DMEM/甘油-80℃保存组、纯甘油4℃保存组(保存1、3个月),应用免疫组织化学方法,检测HGF、bFGF在各组羊膜中的表达。结果:HGF、bFGF主要分布于羊膜上皮层。HGF在DMEM/甘油组和纯甘油组保存1、3个月时的表达明显低于新鲜羊膜组,统计学上差异有显著性(P<0.0001),bFGF在两组保存3个月时的表达明显低于新鲜羊膜组,统计学上差异有显著性(P<0.01)。结论:羊膜经DMEM/甘油-80℃、纯甘油4℃保存后,HGF、bFGF减少,并随保存时间的延长活性逐渐下降。 Objective :To study the expressions of hepatocyte growth factor (HGF) and basic fibroblast growth factor (bFGF) in fresh and two preserved amniotic membranes (AM), and to explore their biological vitality. Methods:The amniotic membranes from the healthy puerperia with abdominal delivery were randomly divided into three groups: fresh group, DMEM/glycerol preserved group at -80℃ and glycerol preserved group at 4℃ (for 1 and 3 months). [mmunohistochemical method was used to investigate the changes in the expressions of HGF and bFGF among the three groups. Results : HGF and bFGF were distributed in the epithelium of AMs mostly. The expressions of HGF in both preserved AMs for 1 and 3 months decreased significantly compared with fresh AM (P〈 0. 0001 ), and the expressions of bFGF in both preserved AMs for 3 months decreased significantly compared with fresh AM (P 〈0.01 ). Conclusion : HGF and bFGF both decrease in AMs preserved in DMEM/glycerol at - 80℃ and in glycerol at 4℃, and their vitality declines gradually with preserving time.
