^(131)I-GM-CSF与^(131)I-anti-CD45分别作用于HL60细胞的比较

摘要 目的:为了比较相同放射性浓度下的131I-GMCSF和131I-anti-CD45对HL60细胞的细胞毒作用,诱导凋亡作用及对细胞周期的影响。我们采用体外放射免疫导向治疗(RIT)模型,用MTT法比较131I-GMCSF和131I-anti-CD45对HL60细胞的细胞毒作用,Annexin V/PI双染法测两处理组的早期凋亡细胞,并用流式细胞术检测其对细胞周期的影响。结果显示131I-GMCSF和131I-anti-CD45对HL60细胞的细胞毒作用非常接近,当放射性浓度≥18.5×108Bq/L时两处理组的HL60细胞存活率都显著下降,两组都能明显诱导HL60细胞凋亡,能显著引起HL60细胞周期发生G2/M期阻滞,两者相比没有显著性差异(P>0.05)。结论:用GM-CSF作为载体代替通常用的鼠型单克隆抗体用于急性髓性白血病(AML)的放射免疫导向治疗是可行的,具有重要的临床意义。 Objective :To compare the cytotoxicity,apoptosis and cell cycle changes of HL60 cells treated with ^131I-GMCSF and ^131I- anti - CD45. Methods :An in vitro radioimmunotherapy (RIT) model was employed. Data were collected from tetrazolium microculture cellar cytotoxicity test (MTT) and flow cytometric analysis. Results:The results showed that ^131I- GMCSF and ^131I- anti -CD45 both induced apoptosis on HL60 cells, and the cells growth inhibition rates were close between the two groups. G2/M arrest was observed in two groups. There was no notable difference between ^131I- GM - CSF group and ^131I- anti - CD45 group( P 〉 0.05 ).Conclusion :It is feasible to treat AML by using GM-CSF instead of mouse monoclonal antibody combined ^131I.
