摘要 目的开发Windows操作系统下基于双波长荧光测钙原理的荧光显微测钙系统软件。方法通过分析单色光源的工作原理得到输出波长和电压之间的函数关系,并以此作为设计软件算法的依据;采用编程框架(MFC)中的应用程序框架和多线程技术,用VC++6.0开发Windows下的应用软件。结果在PC机上实现了荧光显微测钙软件,并用该软件、PC机、单色光源、荧光显微镜、光电倍增管以及数据采集器构建的荧光测钙系统,实现了单个活体β细胞的胞内游离钙离子浓度的测量,测量结果在正常生理范围内。结论软件能够准确测量出活体细胞内游离钙离子浓度,为细胞生物物理研究提供了一种有效的工具。 Objective To design software based on the double-wavelength fluorescence microscopy to measure cytosolic free calcium( [ Ca^2+ ]i) in living cell. Method Through analyzing the work principles of monochromator, the function relation between output wavelength and voltage was obtained. Based on the analysis results as the arithmetic of the software, the application was developed in the integration development environment of VC ++ 6.0, by using the framework of Microsoft Foundation Class Library(MFC) and multithread technology. Result The authors has realized the software and constructed a fluorescence microscopy system using the software, PC, monochromator, fluorescence microscope, MPT and data acquisition to measure the [ Ca^2+ ]i in single living β cell. Conclusion Using this software, the [ Ca^2+ ]i in living cell can be detected. It has been proved to be an effective tool for the research in cellular biophysics.
