摘要
①目的探讨急性期脑梗死病人外周血中性粒细胞(PMN)与低氧-复氧(H/R)血管内皮细胞黏附性,以及抗ICAM-1单抗对其的影响。②方法培养的人类脐静脉内皮细胞株ECV-304经(或不经)H/R和抗ICAM-1单抗处理后,分别加入脑梗死病人外周血PMN并检测黏附率。③结果ECV-304经H/R处理后与脑梗死病人外周血PMN黏附率明显增高;抗ICAM-1单抗可显著抑制脑梗死病人外周血PMN与H/RECV-304黏附。④结论脑梗死病人外周血PMN活化,H/R使血管内皮细胞与PMN黏附性进一步增强;ICAM-1参与介导脑梗死后PMN与血管内皮细胞黏附,抗ICAM-1单抗可起部分阻断作用。
Objective To investigate the adhesion of polymorphonuclear neutrophils (PMN) in patients with cerebral infarction (CI) to hypoxia/reoxygenation (H/R)-stimulated human umbilical vein endothelial cells (ECV-304), and to define the effects of antibodies to intercellular adhesion molecular-1 (ICAM-1 mAb, anti-CD54 antibodies) upon the adhesion. Methods The ECV-304 was treated with (or without) H/R and anti ICAM-1 mAb, and added to the PMN of the CI patients. The adhesion rate was determined. Results Compared with non-H/R-stimulated endothelial cells, adhesion of PMN from CI patients to H/R-stimulated ECV-304 increased highly, but the adhesion was reduced by anti CD54 antibodies, Conclusion PMN is involved in cerebral injury of the CI patients and the adhesion to H/R-stlmulated endothelial cells is enhanced. Adhesion of PMN to endothelial cells is mediated by ICAM-1(CD54), and anti-CD54 antibodies can partially block the adhesion.
