公鸡精子冷冻保存后活率和顶体破损率的相关关系 被引量:4

The Correlation between the Survival Rates and Broken Acrosomic Rates of Cock Spermatozoa

摘要 用BPSE为稀释液,以终浓度为11%(V/V)的甘油作低温保护剂,冷冻洪山公鸡精液。测定了细管型和颗粒型2种类型的冷冻精液解冻后的精子存活率和顶体破损率,并对冻后存活率和顶体破损率进行了相关和回归处理。测定结果表明,细管型冷冻精液解冻后的精子存活率和顶体破损率分别为0.451±0.056和0.392±0.054(n=20)。颗粒型冷冻精液解冻后的精子存活率和顶体破损率分别为0.339±0.040和0.543±0.052(n=20)。前者解冻后的活率极显著高于后者,而顶体破损率则极显著低于后者。相关分析结果表明,细管型冷冻精液解冻后精子存活率与顶体破损率之间的回归方程为y=0.7293-0.7126x,相关系数r为-0.697(P<0.01),而颗粒型冷冻精液解冻后的精子存活率与顶体破损率之间的相关系数为-0.0625(P>0.05),表明两者之间无显著相关。本研究为输入相等数量的活精子时细管型冷冻精液受精率显著高于颗粒型冷冻精液的试验结果提供了一定的理论解释,也为将保存家畜冷冻精液的方法应用于家禽方面作了有益的探索。 With BPSE as extender and glycerol ( with a final concentration of 11%, V/V) as cryoprotectant, semen from Hongshan cock was frozen in both pellets and straws. The post thawed spermatozoa survival rates, broken acrosomic rates of the two types of frozen semen were tested, and the correlation between the survival rates and broken acrosomic rates were analysed. The testing results showed that the average survival rates and broken acosomic rates of post-thawed spermatozoa in straws were 0. 451±0. 056 and 0. 392±0. 054 respectively (n=20), those in pellets were 0. 339±0. 040 and 0. 543±0. 052 (n=20), respectively, t test results showed that the survival rates of the former were significantly higher than the latter (P〈0. 01), but the broken acrosomic rates were significantly lower than the latter(P〈0. 01). Correlationship analysis showed that there was a significantly negative correlationship (-0. 697, P〈0.01) between the survival rates and broken acrosomic rates in straws, with a regression equation of y=0. 729 3--0,712 6x (y, the survival rates of the post-thawed spermatozoa; x, the broken acrosomic rates of the post-thawed spermatozoa ), that in pellets was not significant (-0. 062 5, P〉0. 05). This study to some extent explained the fact that when the same amount of live spermatozoa stored in straw and pellet used the fertilities of the former was significantly higher than the latter, it also provided some benefits for the cryopreservation of pouhry's spermatozoa as an effective method for the conservation of germ plasm in poultry.
