摘要 ①目的通过检测人乳癌组织中组织蛋白酶D(CathepsinD)、MMP2和MMP9三种蛋白酶的表达,探讨其在乳癌演进和异质化过程中的改变及意义。②方法采用免疫组织化学方法,检测58例人乳癌标本、5例正常乳腺标本及5例良性增生乳腺组织标本中三种酶的表达。③结果CathepsinD、MMP2和MMP9在乳癌的肿瘤细胞和间质细胞中均有表达,在肿瘤细胞中呈异质性过表达,而良性增生乳腺组织呈阴性或弱表达;CathepsinD阳性表达与瘤细胞分化程度(分级)有关(χ2=8.35,P<0.05);而MMP2和MMP9阳性表达与淋巴结转移有关(χ2=7.66、17.67,P<0.05)。④结论乳癌CathepsinD、MMP2和MMP9的表达与正常或良性增生乳腺组织有质的差别,是乳癌不断演进和异质化的形态和功能特征之一,与乳癌癌细胞的侵袭和转移能力密切相关。 Objective To investigate the expression of Cathepsin-D, matrix metalloproteinase-2 (MMP 2)and matrix metalloproteinase-9 (MMP-9) in human breast cancer and explore their changes during the progression and heterogeneity of the tumor. Methods Using immunohistochemistry, the expressions of these three proteinases in 58 cases of breast cancer and 10 cases of benign breast samples were examined. Results Cathepsin-D, MMP 2 and MMP-9 were all over expressed in breast cancer heterogeneously compared to benign breast tissue. They were expressed in both cancer tissue and matrix cells, Cathepsin-D was mainly expressed in the ceils which were poorly differentiated (x^2=8.35, P〈0.05), while MMP-2 and MMP 9 were more expressed in tumor tissue which had positive lymph nodes (x^2=7.66,17.67; P〈0.05). Conclusion The expression of Cathepsin-D, MMP-2 and MMP-9 is one of the important character of progression and heterogeneity of the breast carcinoma,and they are associated with the invasion and metastasis of the tumor.
