摘要
①目的研究胃癌组织中由核DNA编码的琥珀酸脱氢酶(SDHD)基因位点的改变情况。②方法选取与SDHD基因紧密连锁的3个微卫星D11S5017、D11S1347和D11S1328,通过PCRSSLP的方法检测了35例新鲜胃癌组织中3个微卫星位点的杂合性丢失(LOH)情况。③结果胃癌组织中,D11S5017位点LOH检出率为12.0%,D11S1347位点LOH检出率为10.0%,D11S1328位点LOH检出率为14.3%。3个位点LOH总检出率为19.4%。④结论在胃癌组织中存在着在SDHD基因LOH的现象,说明了它可能是胃癌中的一种特殊类型的肿瘤抑制因子。
Objective To study the gene alteration of SDHD coded by nuclear DNA in gastric carcinoma. Methods The LOH of three microsatellites-D11S5017, D11S1347 and D11S1328 that closely surrounded the SDHD gene were examined in 35 cases of fresh gastric carcinoma with PCR LOH analysis(SSLP) method. Results In gastric carcinoma, the LOH rate of D11S5017, D11S1347 and D11S1328 was 12.0%,10.0% and 14.3%, respectively. The total rate of LOH in the three sites was19.4%(6/31). Conclusion LOH of SDHD genes exist in gastric carcinoma, which suggests that it can be considered as a special type of tumor suppressor coded by nuclear genes.
