银杏叶提取物对培养中的胎儿血管平滑肌细胞Bcl-2蛋白表达量的影响

摘要 目的观察银杏叶提取物(Egb)对培养中的胎儿血管平滑肌细胞(VSMC)凋亡相关蛋白Bcl-2的影响。方法采用酶消化法结合组织贴块法培养VSMC。加不同浓度Egb干预不同时间段,形态学观察凋亡现象,利用Annexin-V结合PI,流式细胞仪技术检测细胞凋亡率,检测Bcl-2蛋白的表达量变化。结果与对照组相比,随着时间的延长,Egb浓度的增加,细胞凋亡率增加,细胞Bcl-2蛋白表达率明显减少。结论Egb可以诱导培养中的胎儿VSMC凋亡,并可以抑制VSMC凋亡相关蛋白Bcl-2的表达,并且这两种作用呈时间及浓度依赖性。 Objective To investigate the influence of Egb on the express of Fas protein of the VSMCs in culture. Methods Culture VSMCs from the thoracic aortas of the aborted fetus by Collagenase Ⅳ digested combined with tissue anchored.Add uptitratal concentrations of Egb(0μg/ml、200μg/ml、400μg/ml、800μg/ml) to the culture medium of cells which grew well at 5 -8 passages. At the different time point(6h, 12h, 24h), The ratio of apoptosis cells were detected with annexin-Ⅴ combined PI by flow cytometry, then Bcl-2 were detected, which were correlated protein with apotosis by flow cytometry. Results Along with the increase of concentration and exposed time, the apoptosis rate was increased(P =0.00), and along with the increase of Egb concentration and the prolong of exposed time, the expression of Bcl-2 was increased( P = 0. 001 ). Conclusions Egb may induce apoptosis of VSMC. Along with the increased of Egb concentration and exposed time, the apoptosis rate is increased. With the gain of apoptosis rate, the expression of Bcl-2 increased.
