摘要
目的:观察洛伐他汀(LOV)对HL-60细胞Fas抗原表达的影响,探讨其抑制细胞增殖、诱导细胞凋亡的可能分子机制。方法:采用流式细胞仪检测HL-60细胞的细胞周期相分布,细胞凋亡率,Fas抗原表达水平。结果:4、8、16μmol/LLOV处理HL-60细胞48h后,G0/G1期细胞增多、细胞凋亡百分率增加、增殖指数降低、Fas抗原的平均荧光强度增加,与剂量呈正相关。结论:LOV对HL-60细胞有增殖抑制和诱导凋亡作用,上调节Fas抗原表达可能是其分子机制之一。
Objective: To study the effects of lovastatin (LOV) on the expression of Fas antigen and proliferation and apoptosis of HL - 60 ceils, and its possible molecular mechanisms. Methods : Flow cytometry (FCM) was used toanalyse the ceil cycle distribution, the apoptotic rate and the expression of Fas antigen of HL -60 cells. Results: After treatment with LOV at dosages of 4, 8, 16 μmol/L for 48 h, the ceils of G0/G1 phase, the apoptotic rate and the expression of Fas antigen of HL - 60 cells were increased, the proliferation index (PI) of HL - 60 ceils was decreased, these effects of LOV were in a dose dependent manner. Conclusion: LOV inhibit HL -60 cells proliferation and induce the ceil apoptosis, in which, its effects of up - regulating Fas antigen expression in HL-60 cells may play a key role.
