斑点ELISA检测鸡传染性支气管炎病毒及抗体

ESTABLISHMENT OF INDIRECT DOT-ELISA TO DETECTANTIGENS OF AVIAN INFECTIOUS BRONCHITSVIRUS(IBV)AND ANTIBODIES AGAINST IBV

采用鼠抗鸡传染性支气管炎病毒（ＩＢＶ）高免血请作第１抗体，利用酶标羊抗鼠ＩｇＧ作第２抗体，建立间接法Ｄｏｔ－ＥＬＩＳＡ程序检测ＩＢＶ抗原；利用鸡抗ＩＢＶ血清阻断试验检测ＩＢＶ抗体。试验表明，本程序检测ＩＢＶ抗原及其抗体具有高度过感性和特异性。最低抗原检出量达１０￣（－８）ｇ数量级，阳性检出率９８％，阳性阻断率１００％。

In this experiment,standard Indirect Dot-ELISA process was established to detect antigens ofIBV and Antibodies against IBV. The results demonstrated that 1. 64×10￣(-8)g was the least amount of puri-fied IBV viral antigens that the method could detect under the optimal conditions.It's specificity in the detec-tion of IBV antigens has been well corroborated by the blockage test of IBV positive chicken serum, and thecross reaction test for the detection of IBV, IBDV, 10O SPF chicken embryos infected with C_(9001) strain,andIt's allantoic fluid was detected by the Indirect Dot-ELISA. The positive percentage of the test was 98%, 147serum of ' AA' chicken,infected with C_(9001) strain were used to block the Indirect Dot-ELISA,the resultsshowed that the percentage of blockage test was 100%.
