导入人端粒酶逆转录酶基因致人外周血淋巴细胞长期生存和增殖的研究

摘要 用外源性人端粒酶逆转录酶(hTERT)基因导入正常人外周血淋巴细胞,延长细胞生存期并增强其增殖能力。将含有外源性hTERT基因的质粒导入人外周血淋巴细胞,RT-PCR检测hTERTmRNA在细胞内的表达,间接免疫荧光检测细胞内hTERT蛋白,并观察转染后细胞的生物学特征。结果:RT-PCR检测到转染后细胞内hTERT基因在mRNA水平稳定表达以及胞内存在的hTERT蛋白,细胞生存期延长,增殖旺盛,细胞生物学特征正常。因此,外源性hTERT基因表达可使正常人外周血淋巴细胞生存期延长、增殖旺盛,并保持正常细胞生物学特征。
