摘要
目的研究牛膝总甾酮在小鼠体内的药物动力学。方法以蜕皮甾酮为指标,用HPLC法测定给药后小鼠血清中总甾酮的浓度。结果小鼠口服给药后5min血清中即可检出蜕皮甾酮,总甾酮在小鼠体内呈一室模型分布,其主要动力学参数为:A=0.3024μg.ml-1;Ke=0.0014min-1;Ka=0.3520min-1;Lagtime=0.3318min-1;t1/2(ka)=1.9692min;t1/2(ke)=488.6178min;Cmax=0.2945μg.ml-1;Tpeak=15.7284min;AUC=212.2944μg.min.ml-1;Cl/F(s)=942.0870ml.min-1;V/F(c)=664.1025×103ml。结论HPLC法适用于牛膝总甾酮血药浓度的测定,总甾酮在小鼠体内吸收很快。
OBJECTIVE To study the phannacokinetic characters of total phytosterone from Achyranthes bidentata in mice. METHODS HPLC method was adopted.RESULTS The results indicated that the total phytosterone was detected after po total phytosterone 5 min. The sertma concentration - time curve conformed to one - eompartment open model. The Pharmaeokinetic parameters were A =0. 3024μg·ml^-1; Ke=0.0014min^-1 ; Ka=0. 3520min^-1 ; Lag time = 0. 3318min^-1 ; t1/2 (ka) =1. 9692min; t1/2 (ke) = 488. 6178min; C = 0. 2945μg·ml^ -1 ; Tyrok=15. 7284min; AUC = 212. 2944μg·min·ml^-1 ; Cl/F(s)=942. 0870ml·min ^-1 ; V/F(e) = 664. 1025×10^3ml. CONCLUSION HPLC is suitable for determining total phytosterone in mouse serum. The absorption of total phytosterone is very fast.
